TRADD, which stands for Tumor necrosis factor receptor type 1-associated DEATH domain protein, is a protein involved in the regulation of cell death and immune response pathways. It plays a vital role in the activation of apoptotic signaling and the fulfillment of various functions within the immune system.
TRADD is a protein adapter that is primarily found associated with the cytoplasmic tail of tumor necrosis factor receptor 1 (TNFR1). Upon activation of TNFR1 by binding to tumor necrosis factor (TNF), TRADD binds to the receptor and recruits other proteins, such as FADD (Fas-associated protein with death domain), to form a multiprotein complex known as a signaling complex. This complex leads to the activation of downstream signaling pathways involved in cell death, such as apoptosis.
Apart from its role in mediating apoptosis, TRADD also participates in the regulation of NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) signaling, which is involved in immune and inflammatory responses. TRADD interacts with other proteins, such as TRAF2 (tumor necrosis factor receptor-associated factor 2) and RIP (receptor-interacting protein) kinases, to activate the NF-κB pathway.
